Glucagon is a peptide consisting of 29 amino acids produced by the alpha cells of the islets of
Langerhans.Glucagon has been established as an important hormone that orchestrates glucose,lipid,and amino acid metabolism,as well as energy intake and expenditure.
Glucagon is a counter-regulatory hormone opposing the actions of insulin in glucose homoeostasis. Glucagon is thought to play an important role in the maintenance of fasting and postprandial glucose homeostasis.By stimulating hepatic glucose output, glucagon counterbalances the action of insulin and serves to maintain circulating glucose and prevent insulin mediated hypoglycemia.

Both type 1 and type 2 diabetes frequently exhibit an inappropriately high glucagon response to a meal.The high levels of glucagon have been shown to contribute importantly to diabetic hyperglycemia and can result in ketoacidosis.Relative hyperglucagonemia, in the setting of deficient insulin secretion may contribute to the development of fasting and postprandial hyperglycemia in the patients.

Measuring glucagon – not so simple!
l While glucagon’s actions may be unique, its sequence is not. Herein lies a major part of the measurement quandary that has faced scientists in years past.
l Circulating levels of homologous proteins should also be taken into consideration and can make cross-reactivity in a glucagon assay particularly problematic, which is especially true for glicentin.
l Inaccurate glucagon readings can have enormous negative implications on research and clinical decisions.
l A true insulin:glucagon ratio cannot be determined if using assays (for either biomarker) that are not highly specific.
